N-棕榈酰乙醇胺对大鼠抑郁样行为的研究


摘 要
 
目的:探究N-棕榈酰乙醇胺(N-palmitoylethanolamide,PEA)对大鼠抑郁样行为的影响,揭示其抗抑郁的机制。
 
方法:取健康雄性SD大鼠50只适应性饲养5 d,依据体重指标将合格动物随机分为5组,10只/组:空白组(normal control,NC)、模型组(model control,MC)、氟西汀组(fluoxetine,FLU,10 mg/kg,po)、PEA组(10 mg/kg,po)和拮抗剂组(PEA 10 mg/kg,po + MK886 3 mg/kg,ip)。NC 组10只一笼群养,其余4组均接受独笼饲养。造模过程中,每周记录大鼠体重,第28 d进行蔗糖水偏好实验(sucrose preference test,SPT)和新环境进食抑制实验(关键词:N-棕榈酰乙醇胺;慢性不可预见性应激;抑郁;大鼠
 
Abstract
 
Objective: To explore the effect of N-palmitoylethanolamide (PEA) on depression-like behavior in rats and to reveal its anti-depressant mechanism.
 
Methods: Take healthy male SD rats 50 adaptability raised 5 d,according to the weight indicators qualified animals were randomly divided into 5 groups, 10 / groups: blank group,normal control, NC), model group, model control, MC), the fluoxetine group (fluoxetine, FLU, 10 mg/kg, Po), PEA group (10 mg/kg, Po) and antagonist group (PEA 10 mg/kg, Po + MK886 3 mg/kg, IP). Ten rabbits in NC group were raised in a group cage,and the other 4 groups were raised in a single cage. During modeling, the body weight of rats was recorded every week,and sucrose preference test (SPT) and novelty feeding test (NSFT) were conducted on the 28th day. After modeling, the protein expressions of synaptophysin (SYN) and postsynaptic density protein95 (PSD95) were detected by Western blot (WB).
 
Results: In the behavioral experiment, the basal body weight of the rats was similar (P<0.05).On the 28th day of the experiment, compared with the NC group, the body weight,sucrose preference rate and feeding latency of rats in MC group were significantly decreased (P<0.01), and the sucrose preference rate was significantly decreased (P<0.001), and the feeding latency was significantly increased (P<0.001), indicating the success of modeling.Compared with MC group,the body weight and sucrose preference rate of rats in Flu and PEA groups were significantly increased (P<0.01, P<0.01), and the feeding latency was significantly decreased (P<0.01, P< 0.05), indicating that Flu or PEA can improve the depression-like behavior of rats.Compared with the PEA group,the combined use of PEA and MK886 resulted in a significant decrease in body weight (P<0.05), a significant decrease in sucrose preference rate (P<0.01), and a significant increase in feeding latency (P<0.05), indicating that the behavioral improvement effectof PEA on rats was partially or completely cancelled. In western blotting assay, compared with NC group,the protein contents of SYN and PSD95 in MC group were significantly decreased (P<0.001, P<0.001).Compared with MC group,the protein contents of SYN and PSD95 in Flu and PEA groups were significantly increased (P<0.001, P<0.001), suggesting that Flu or PEA can promote the growth and development of neurons.Compared with PEA group,the protein contents of SYN and PSD95 in PEA+MK886 group were significantly decreased (P<0.001, P<0.001), suggesting that MK886 antagonized PEA.
